美拉諾坦 II

美拉諾坦 II（Melanotan II）[1]是一種人工合成的α-黑素細胞刺激激素（α-MSH）類似物，α-MSH屬於肽類激素。

本品在開發期間的目標是作為治療女性性功能障礙和勃起功能障礙的候選藥物，但在2003年停止開發。截至2018年，尚無含有美拉諾坦 II 的產品上市，有關此藥物的藥物開發也都已經停止[1]。 在網路上可以發現許多標榜含有美拉諾坦 II 的無牌商品在市場上流通，被認為是有效的仿曬劑，但有成色不均勻、長新的痣或原有的痣變黑或變大等副作用，因此醫療機構不建議使用[2][3]。

作用機轉

美拉諾坦 II 為黑皮質素受體的非選擇性激動劑，可活化MC1、MC3、MC4、MC5等受體[4]。

美拉諾坦 II 能夠促進黑素細胞的生成，可能是藉由活化MC1受體所致，而其在性方面的效果被認為與其激活MC4受體的能力有關（MC3 受體可能也與其有關）。[5][6]

美拉諾坦 II 的不良反應包括潮紅、噁心、嘔吐、伸展、哈欠，和食欲不振（食慾不振與MC4的活化相關）[7][8]。

歷史

在1960年代初的研究發現，給予大鼠α-MSH會引起其性反應。到1980年代在許多實驗室中仍繼續做這樣的實驗，亞利桑那大學的科學家開始試圖開發α-MSH或其類似物作為潛在的仿曬劑，並合成和測試了幾個類似物，包括美拉諾坦 I 和美拉諾坦 II[9][10]。

在該藥物研發的起步階段，其中一位科學家決定拿自己做實驗，將兩倍預期劑量的美拉諾坦 II 注射到自己身上，結果造成他連續勃起8小時，並伴有噁心和嘔吐[11]。

於是亞利桑那大學運營的技術轉讓公司 Competitive Technologies 便將美拉諾坦 I以仿曬劑候選物的形式技轉給一家名為Epitan的澳大利亞新創公司（2006年更名為Clinuvel）[12][13]。而美拉諾坦 II 則以性功能障礙藥物被技轉給帕拉丁科技（Palatin Technologies）[10]。帕拉丁技術公司在2000年結束了美拉諾坦 II 的開發，重新開發另一種專利新藥，即布美諾肽（可能為美拉諾坦 II的代謝物），與美拉諾坦 II 的不同處在於布美諾肽以一個羧基取代美拉諾坦的酰胺基[6][14]。於是 Competitive Technologies 控告帕拉丁科技違約，並聲稱其具有對布美諾肽的權利[15]。雙方於2008年和解，帕拉丁科技保有布美諾肽的權利，並支付80萬美元給 Competitive Technologies ，而 Competitive Technologies 則繼續保有美拉諾坦 II 的權利[16]。

社會和文化

許多產品在市場上銷售，在健身房和美容院中被稱為“美拉諾坦 ”或“美拉諾坦 -1”或“美拉諾坦 -2”。[17][18] [19]
